Depuis les origines de la civilisation, Apocalypse (Oscar Isaac), le premier et plus puissant mutant du monde, a absorbé de nombreux pouvoirs, devenant à la fois immortel et invincible, adoré comme un dieu. Se réveillant après un long sommeil de plusieurs milliers d'années, désillusionné par le monde qu'il découvre, il réunit une équipe de puissants mutants, dont Magneto (Michael Fassbender) pour nettoyer l'humanité et créer un nouvel ordre, sur lequel il règnera. Raven (Jennifer Lawrence) et Professeur X (James McAvoy), à la tête d'une équipe de jeunes X-Men, vont devoir joindre leurs forces pour arrêter leur plus dangereux ennemi et sauver l'humanité toute entière d'une destruction totale.
